        Top-Down Characterization of a Site-specific Antibody-drug Conjugate

        Peptide mapping is the gold standard for in-depth characterization of biotherapeutics; however, sample preparation, LC runtime and data analysis can be labour intensive and time consuming. Top or middle-down mass spectrometry experiments show promise as an alternate technique for primary order structure confirmation. However, historically top or middle-down characterisation has been limited by bottlenecks in data processing, and lower sequence coverage.

        In this webinar, we show how rapid high sequence coverage of mAbs and ADCs can be achieved with advanced top-down fragmentation capabilities such as Higher-energy Collision Dissociation (HCD), Electron Transfer Dissociation (ETD) and Ultra-Violet Photo-Dissociation (UVPD). In addition, we show that the technique can pinpoint sites of oxidation resulting from oxidative stress. Finally, learn about advances in automated software processing that easily map the protein structure and provide confirmation of sequence coverage and process induced modifications.

        Dr. Oscar Hernandez Alba is currently working as a postdoctoral researcher in the LSMBO laboratory at the University of Strasbourg. He obtained his PhD degree at the University of Paris-Sud under the supervision of Dr. Philippe Maître. He joined the group of Pr. Bela Paizs during his first postdoctoral stay at Bangor University in Wales, United Kingdom. His research domains are: mass spectrometry, native mass spectrometry, infrared spectroscopy, ion mobility, and the study of mAbs, ADCs and protein-protein interactions from the intact to middle-down level characterization.

        Dr. Stephane Houel was educated in environmental geochemistry with a Ph.D. in Environmental Sciences from the University of Quebec in Montreal and focused on lignin biomarkers in hydroelectric reservoirs. Mass Spectrometry is a common language across multiple disciplines, and Stephane had the opportunity to apply it in anti-doping, oceanography, limnology, proteomics and protein biotherapeutics analysis. Prior to joining Thermo Fisher Scientific in 2015, Stephane worked for another mass spectrometer vendor in a marketing group focusing on the biopharmaceutical industry. Recently, Stephane has focused on top-down applications using an Orbitrap Fusion Lumos Tribrid mass spectrometer.
